The 25-year-old was driving a ute along the Calder Alternate Highway about noon on Tuesday when he crossed onto the opposite side of the road and collided head on with a truck.
The man, the only occupant of the vehicle, died at the scene. The truck driver suffered minor injuries and experienced shock.
Acting Senior Sergeant Bruce Simpson from Bendigo Highway Patrol said an exact cause of the crash was yet to be determined but early indicators suggested fatigue had played a part.
"We have spoken to several witnesses. At the moment it is too early to establish an exact cause," Acting Senior Sergeant Simpson said.
"At this stage the indications are the deceased driver has crossed over onto the wrong side of the road."
Acting Senior Sergeant Simpson said drivers needed to take care every time they started up their vehicles.
"We can't reiterate this message enough. Just be careful on the roads," he said.
"Make sure you don't drink or drive, you pay attention to what is going on (and) you are well rested.
"And at the moment, with school holidays on, there are a lot of families on the roads and we don't want them to come into harms way."
Acting Senior Sergeant Simpson said Tuesday's crash scene was confronting for all involved.
"The amount of emergency services involved is enormous," he said.
"And the deceased is someone's family, it's distraught for everybody."
Acting Senior Sergeant Simpson said several witnesses were helping police with their investigations.
The road toll sits at 184, compared with 175 at the same time last year.
Anyone who witnessed the collision is asked to contact Crime Stoppers on 1800 333 000 or to make a confidential report at www.crimestoppersvic.com.au
